چکیده :

A 1342 bp fragment, AFHA, was amplified by PCR from Aeluropus littoralis genomic DNA and its nucleotide sequence was determined. The sequence analysis showed that the fragment was amplified from a gene that codes for a plasma membrane H+-ATPase. The primers were designed based on the highly conserved sequence of plant plasma membrane H+-ATPases. The nucleotide sequence of AFHA was compared with the sequence of Alha1, another gene encoding a plasma membrane H+-ATPase isoform of A. litoralis. The results showed that AFHA was amplified from a new isoform that has nearly 97% similarity with Alha1. The comparison of predicted amino acid sequences of these isoforms revealed that they are different in 8 of the 303 amino acid sites. The codon usage analysis of these isoforms showed that these two isoforms differ in 17 of the 33 codon usage, but around half of them resulted in a synonymous substitution. Since, these isoforms were isolated from the plants growing in different regions, two hypotheses may be considered. The first hypothesis is that the two sequences correspond to one locus which has gone through two different evolutionary paths because of geographical separation and the second one considers them as two different loci that were derived from a common ancestral locus due to duplication event.
