Protein A is a 42 kDa surface protein originally found in the cell wall of the bacterium Staphylococcus aureus. It is encoded by the spa gene. It has found use in biochemical research because of its ability to bind immunoglobulins. It binds proteins from many of mammalian species, most notably IgGs. Protein A is used to purify, immobilize or detect immunoglobulin. It is also used for immunoprecipitation or immunoadsorption-based assays. Immunoprecipitation studies with protein A conjugated to beads are also commonly used to purify proteins or protein complexes indirectly through antibodies against the protein or protein complex of interest. In this project the gene encoded Protein A along with its signal peptide and its five IgG-binding domains was cloned in Bacillus subtilis. It was also secreted to the culture medium after expression and in this way the purification of the protein would become easier.
