Esterase and hemolytic Activities in Various Candida Species isolated from the oral cavity of the diabetic patients

Introduction: Diabetes mellitus as a chronic metabolic disease was appeared in patients with partial or complete deficiency of insulin secretion or disorder in action of insulin on tissue. Diabetes melitus prepares conditions for more growth of candida. Candida tends to cause candidiasis due to have plenty virulence factors such as esterase, hemolysin , phospholipase . The aim of this study is comparison of Esterase and hemolytic Activity in Various Candida Species isolated from the oral cavity of the diabetic patients and non-diabetic individuals. Materials & methods: From mouth of 95diabetic patients (35 men and 60 women) and 95 normal persons (42 men and 53 women) were sampled. Yeast species which grow on Sabouraud dextrose agar were identified based on germ tube test, culturing on CHROMagar Candida medium, corn meal agar and ability to grow at 45°. Hemolysin activity was evaluated with a blood plate assay and esterase activity was assayed by using of the tween 80 opacity test . Results: Various species of candida were isolated from mouth of 60 %( 57 out of 95) of the Diabetic and 25 %( 24 out of 95) of the non-diabetic individuals. Estrase activity was detected from 100% candida species. Only 21.6% of C. albicans isolated from diabetic patient produced esterase enzyme as +3 but others produced it as degree +2 or +1. Hemolitic activity were assayed in C. albicans, C. dubliniensis, C. glabrata and C. krusei as 0.79, 0.58, 0.66 and 0.74 respectively. Hemolytic activity was not significantly different in the two groups of diabetic and non-diabetic. Discussion: The oral carriage of C. albicans in the diabetic group (n=42; 66.7 %) was significantly greater than the control group (n=16; 57.1 %). Esterase enzyme of C. albicans as virulence factor, in diabetic group was higher than that in non-diabetic group. Although C. albicans remains the most frequently isolated yeast pathogen but other species are also involved in the pathogenesis of human.
